چکیده

It is well known that the pineal gland hormone melatonin has a direct effect on pigment dispersion in amphibian melanocytes. This pineal hormone has also been reported to influence pituitary melanocyte-stimulating hormone (MSH) activity in the rat. Melatonin is formed from serotonin (5-hydroxytryptamine) in two metabolic steps: first, serotonin is enzymatically metabolized by N-acetyltransferase to N-acetylserotonin, and second, this is converted to melatonin by hydroxy-indole-O-methyltransferase (HIOMT). The N-acetyl-transferase activity may be induced by stimulation of a j3-adrenergic mechanism. In the present experiment we tested the ability of the p-adrenerg c stimulating drug, isoprenalin (INN, NFN)

Stimulation of C14-melatonin synthesis from C14-tryptophan by noradrenaline in rat pineal in organ culture.

Previous work has shown that the activity of the melatonin-forming enzyme in the rat pineal gland is elevated in rats kept in continuous darkness as compared to those kept in continuous light. Information about environmental lighting reaches the pineal gland via nerves that liberate noradrenaline. Rat pineal glands in organ culture can form C(14)-melatonin from C(14)-tryptophan as follows: tryp...

The Syrian hamster pineal gland responds to isoproterenol in vivo at night.

Failure of isoproterenol (ISO) injections to raise pineal melatonin content has generated doubt about beta-adrenergic control of the melatonin rhythm in Syrian hamsters. However, the effect of ISO injected at night after light-induced reduction of pineal melatonin has not been reported. In this study, light exposure began at 6 1/4 h into one (normally 10-h) dark phase. The hamsters were injecte...

Isoproterenol-stimulated melatonin production by perifused rat pineal glands: age- and time-related effects.

OBJECTIVES The purpose of this study was to investigate and compare the effects of the beta-adrenergic agonist, isoproterenol, on induced increase in melatonin production in the pineal gland of young and old rats, at different circadian stages. MATERIALS AND METHODS We report here the effects of 10(-6) M isoproterenol-stimulated melatonin production by perifused pineal glands obtained from yo...

Inducible cyclic AMP early repressor protein in rat pinealocytes: a highly sensitive natural reporter for regulated gene transcription.

Rhythmic activity of arylalkylamine N-acetyltransferase (AANAT) determines melatonin synthesis in rat pineal gland. The transcriptional regulation of AANAT involves the activating and inhibiting transcription factors of the cyclic AMP (cAMP)-signaling pathway, cAMP response element-binding protein and inducible cAMP early repressor (ICER), respectively. Activation of this pathway is centered ar...

Melatonin in mice: rhythms, response to light, adrenergic stimulation, and metabolism.

There has been relatively little research conducted on pineal melatonin production in laboratory mice, in part, due to the lack of appropriate assays. We studied the pineal and plasma rhythm, response to light, adrenergic stimulation, and metabolism of melatonin in CBA mice. With the use of a sensitive and specific melatonin RIA, melatonin was detected in the pineal glands at all times of the d...
